Synthèse d'anti-androgènes organométalliques non stéroïdiens et application au traitement du cancer de la prostate

Abstract : Prostate cancer is a major societal problem equivalent to that of breast cancer in women. The majority of these cancers are hormone-dependent and one of the commonly used treatments consists in the administration of non steroidal anti-androgens. These molecules inhibit the effects caused by androgens, by competing with the same AR receptor binding site. The commercial non steroidal anti-androgens are flutamide and its active metabolite, 2-hydroxyflutamide, nilutamide and bicalutamide. However, because of the phenomenon of drug resistance that frequently occurs with long-term treatment, the search for more effective molecules becomes imperative. We chose to add a ferrocenyl group on the anti-androgen structure, because it has interesting cytotoxic properties. This research work presents the synthesis of novel ferrocenyl analogues of commercial anti-androgens. Once they are synthesized, they are tested for their relative binding affinity to AR receptor and for their in vitro anti-proliferative activity on prostate cancer cell lines. These biological tests will allow us to know whether these molecules are promising for the treatment of prostate cancer.
