Le transcriptome ABC dans la différenciation, la détoxication et la chimiorésistance.

Abstract : The human ATP-binding cassette (ABC) transporters comprise a large family of membrane transport proteins. Many ABC genes play a role in human genetic disease, and several induce resistance phenotypes. This paper presents the expression pattern of the complete human ABC superfamily by real-time RT-PCR in various organs, or in response to treatment in humans and mice. We studied the effect of colchicine exposure on human cell lines and mice. We have shown that this treatment induce ABC transporters expression. We have established the first mapping of ABC transporters during development liver. Some transporters absent in the fetal liver and speak after birth, probably play a role in the adult liver. By contrast the expression of some transporters extinguished after birth, suggesting a role in hematopoiesis. We measured the expression of ABC transporters in patients with acute leukemias correlated with their sensitivity to chemotherapy. In an interesting way, we have shown that transporeters not described in the resistance could contribute to a poor prognosis. Finally, we studied the ABC transcritpome in human stem cells at different stages of differentiation. Some differences of expression may characterize a state of differentiation and to be involved in self-renewal or differentiation of stem cells.
