Résumé

This thesis has been carried out at the Laboratory for Optics and Biosciences (LOB, Ecole Polytechnic) under the direction of Antigoni Alexandrou. One of the themes of this laboratory is the development and application in the field of biology of a new type of nanometer-sized fluorescent probes. These nanoparticles (NPs) consist of a matrix of yttrium vanadate doped with europium ions (Y1-xEuxVO4) are individually detectable by optical microscopy through the fluorescence they emit. NPs These can be further functionalized to be linked to a biomolecule which one seeks to determine the activity, location or even the dynamics. They join a long line of fluorescent probes which we will briefly describe the various developments. This work has benefited from collaboration with the team Thierry and Jean-Pierre Gacoin BOILOT Laboratory of Condensed Matter Physics (LPMC, Ecole Polytechnique) for the synthesis and functionalization of NPs with the team of Michel Robert Popoff Unit Anaerobic Bacteria and Toxins (TAO, Pasteur Institute) for monitoring and ε toxin with that of Pierre-Louis Tharaux of Cardiovascular Research Centre Inserm Lariboisière for the study of vascular signaling of endothelin- 1.
Ce travail de thèse a été réalisé au Laboratoire d'Optique et Biosciences (LOB, Ecole polytechnique) sous la direction d'Antigoni Alexandrou. Une des thématiques de ce laboratoire est le developpement et l'application dans le domaine de la biologie d'un nouveau type de sondes fluorescentes de taille nanométrique. Ces nanoparticules (NPs) constituées d'une matrice de vanadate d'Yttrium dopée par des ions Europium (Y1−xEuxVO4) sont détectables individuellement en microscopie optique par le biais de la fluorescence qu'elles émettent. Ces NPs peuvent de plus être fonctionnalisées afin d'être couplées à une biomolécule dont on cherche à déterminer l'activité, la localisation ou même la dynamique. Elles s'inscrivent dans une longue lignée de sondes fluorescentes dont nous allons brièvement décrire les différentes évolutions. Ce travail a profité de la collaboration avec l'équipe de Thierry Gacoin et Jean-Pierre Boilot du Laboratoire de Physique de la Matière Condensée (LPMC, Ecole polytechnique) pour la synthèse et la fonctionnalisation des NPs, avec l'équipe de Michel Robert Popoff de l'unité Bactéries Anaérobies et Toxines (BAT, Institut Pasteur) pour le suivi de la toxine ε et avec celle de Pierre-Louis Tharaux du Centre de Recherche Cardiovasculaire Inserm-Lariboisière pour l'étude de la signalisation vasculaire de l'endothéline-1.
