Involvement of HIV in the distribution of dendritic cells between lymphoid organs and blood

Abstract : Dendritic cells are a heterogeneous population of cells of hematopoietic origin. These cells are involved in the innate and adaptive immune response. They can be divided into two groups, myeloid dendritic cells (mDC) and plasmacytoid (pDC). pDC are known to be the main type I interferon producing cells, which have strong antiviral activity. Infection with human immunodeficiency virus (HIV) has an impact on the distribution and function of dendritic cells. The number of dendritic cells is decreased in the blood of patients with HIV. This thesis investigates the mechanisms related to this decrease. In particular the hypothesis of the homing of dendritic cells in lymphoid organs. We show an accumulation of pDC, not mDC in the spleen of infected patients. In the lymph node of cynomolgus macaques in early SIV infection, animal model of HIV infection, we also show a homing of pDC. In the spleen, pDC were found immature and do not seem to produce IFN-α. Other cells such as macrophages and T and B cells appear to responsible of the IFN-α production. In contrast, in monkey, the production of IFN-α in primoinfection seems due to pDC. The IFN-α production correlated with the appearance of mechanisms associated with tolerance as the secretion of indoleamine 2,3-dioygénase and induction of regulatory T lymphocytes. The production of IFN-α in primary infection seems thus necessary to remove the virus. However when the production reaches a high level, it has deleterious effects for the establishment of a sterilizing immune response via induction of regulatory mechanisms. In the chronic stage of infection, the production of IFN-α appears to promote the general activation of the immune system and promote its destruction
