Etudes épidémiologique et phylogénétique chez Bartonella henselae par la technique MLVA

Abstract : The aim of this study was to develop a simple, highly efficient and transferable tool for Bartonella henselae typing, based on multiple Locus variable Number tandem repeat Analysis (MLVA). Five "main" VNTRs were selected according to their polymorphism ( BHV-A, B, C, D and E). Their combination allowed us to evaluate strain diversity and to establish the relationships between different strains of Bartonella henselae. When tested on 178 B. henselae isolates and strains, a diversity index of 0.98 and 99 profils were obtained. MLVA profiles were grouped into 2 main groups named A and B. Group A was exclusively constituted by genotype II feline isolates. Group B included the other feline isolates, one dog isolate and the totality of human isolates (20 human isolates). A study carried out on some cat isolates and their owners showed that MLVA is very efficient for traceability studies. The less polymorphic VNTR seem to constitute geographic markers,while for the most polymorphic VNTR, some alleles were particularly associated to human isolates. There was no common profile between genotypes I and II. Our observations suggest that all group B isolates, i.e. all genotype I isolates (of human and feline origins) and all human isolates (belonging to both genotypes) could be phylogenetically derived from one sub-population of genotype II feline isolates (group A). Moreover, MLVA was efficient for typing "variant" isolates of Bartonella henselae obtained from wild felids. We compared MLVA performances with those of PFGE, MLST and MST. MLVA appears as the most discriminatory technique and is, in addition, more stable than PFGE. Our data suggest that the group B could contain all zoonotic isolates and strains, and that among the 5 intragenic polymorphic VNTR used for MLVA, some of them at least could play a role in their zoonotic potential, in their persistence in cats, and/or in their virulence for humans.
