Abstract : Biophotonics enabled numerous scientific and technological advances for understanding the mechanisms ruling the living as well as in the field of medicine. New tools are regularly developped in order to extend further the limits of the understanding of the molecules constituting the living, and to improve the diagnostics and treatment of diseases. Optical biochips are tools participating into a better understanding of biomolecular surface interactions, and permitted high throughput genetic diagnostics or the simultaneous characterization of a large number of protein interactions in parallel. Furthermore, surface plasmon resonance-based biochips enable the dynamical characterization of biomolecular interactions, without the need of using fluorescent markers The main goal of this work was to develop experimentally a biochip system based on surface plasmon resonance able to dynamically characterize anisotropy resulting from the mean orientation of surface-bound biomolecular assemblies. The system's validation was realized through three applications : the observation of biomolecules orientation effected by a change in the fluidic regime ; the characterization of the anisotropy induced by an electric field on a nanometric-thick biomolecular assembly ; and finally the orientation of strings of micronic magnetic beads under a magnetic field. The second goal of this work was to study and develop new laterally-structured metallic substrates. The local perturbation of the evanescent field propagating at the surface of metal was observed through the use of micro- to nano- scale structured samples.
