Imagerie ultrasonore ultrarapide du cerveau social

Abstract : Ultrasound imaging is a new emerging tool for neurofunctional imaging. Since 2011 and the first proof of concept, numerous preclinical projects were led on various topics such as pathology studies (Alzheimer and schizophrenia for instance), small animal tonotopy study or else social interactions.To study social interactions with this technique, awake imaging is needed. Awake brain functional imaging is possible in ultrasound but it comes withdifficulties not only with the image generation and the experimental setup but also with the interpretation of the results because of the movement artifacts.Being able to perform awake imaging is also very interesting as it allows to get rid of the anesthesia which is known to alter metabolism.Most of the work presented in this document consists in the development of experimental techniques and numerical methodologies to perform functionalultrasound brain imaging (fUS) in awake small animals. The leads that are proposed will be applied on concrete examples with the help of the Pasteurinstitute (study of a model of an autistic trouble) and the Paris institute for neuroscience and psychiatry (social and environmental recognition).fUS is based on the neurovascular coupling: it allows an indirect measure of brain activity by monitoring blood flow variations in the stimulated brain areas.This hemodynamic response is quite slow and thus this technique cannot depict faster transient physiological phenomena. To address this problem, theneuromechanical coupling, that is the link between cerebral activity and tissue stiffness, is investigated for the first time in ultrasound.
